Upstream Synthesis Route
  • To synthesize Potassium (2-methoxyethyl)trifluoroborate (KF(MeOCH2CH2)BF3), one might commence with 2-chloroethanol as the starting material. Firstly, perform an O-alkylation using a methylating agent such as dimethyl sulfate or methyl iodide to obtain 2-methoxyethanol. This alkoxyethanol is then converted to its corresponding tosylate or mesylate ester by treatment with p-toluenesulfonyl chloride or methanesulfonyl chloride respectively in the presence of a base, typically triethylamine (TEA).

Next, conduct a nucleophilic substitution of the tosylate/mesylate with a fluoride source, for instance, tetrabutylammonium fluoride (TBAF) to yield 2-methoxyethyl fluoride. This fluoride is then subjected to a boronation reaction using a boron trifluoride (BF3) donor, such as triethylborane, creating the corresponding 2-methoxyethyltrifluoroborate ester.

Finally, a salt metathesis is carried out by treating the ester with potassium carbonate (K2CO3) in a suitable solvent such as aqueous solution or acetonitrile, affording the desired Potassium (2-methoxyethyl)trifluoroborate product upon purification, commonly executed through crystallization or precipitation, followed by filtration and drying.
